DESCRIPTION:“Powers (and Problems) of Proteases in Tissue Destructive Diseases”\nProfessor\, Biomedical Engineering\nAssociate Chair for Graduate Studies\nGeorgia Institute of Technology \n  \nAbstract: Dr. Platt’s research centers on proteolytic mechanisms of tissue remodeling during disease progression using both experimental and computational approaches. Many of his diseases of focus are health disparities in the U.S.\, but global health concerns: pediatric strokes in sickle cell disease\, HIV-mediated cardiovascular disease\, predictive medicine for breast cancer\, and also COVID-19. His research has taken him to South Africa and Ethiopia for collaborative work to find solutions for low resource settings\, which have broadened opportunity and forced creative solutions. Centered agents are the cysteine cathepsins\, which are the most potent mammalian collagenases and elastases\, but cathepsin pharmacological inhibitors continue to fail human clinical trials\, mostly due to unexpected side effects. This suggests there are underlying regulatory behaviors or feedback loops yet to be elucidated. During this seminar\, Dr. Platt will discuss 1) experimental and computational tools to better quantify and model protease activity\, 2) concept of a proteolytic network in tissue destructive diseases\, and 3) fundamental insights and consequences of the underlying enzymology to improve pharmacological targeting and application for multiple diseases.           \n   \nBio: Dr. Manu Platt received his B.S. in Biology from Morehouse College in 2001 and his Ph.D. from the Georgia Tech and Emory joint program in biomedical engineering in 2006. He finished his postdoctoral training at MIT prior to returning to Georgia Tech and Emory in the joint department of Biomedical Engineering in 2009.  His research centers on proteolytic mechanisms of tissue remodeling during disease progression using both experimental and computational approaches. These diseases of focus are health disparities in the U.S.\, but global health concerns: pediatric strokes in sickle cell disease\, personalized and predictive medicine for breast cancer\, and HIV-mediated cardiovascular disease\, which has taken him to South Africa and Ethiopia for collaborative work to find solutions for low resource settings. His work has been funded by NIH Director’s New Innovator Award\, International AIDS Society\, Georgia Cancer Coalition\, and the National Science Foundation. Integrated with his research program are his mentoring goals of changing the look of the next generation of scientists and engineers. DESCRIPTION:“New insights into aneuploidy in mammalian embryos”\nMelina Schuh\, Ph.D.\nDirector\, Department of Meiosis\nMax Planck Institute for Biophysical Chemistry \nAbstract: The Schuh lab studies meiosis in mammalian oocytes. In her presentation\, Melina will summarize her lab’s recent research on the spindle in mammalian oocytes. Her lab found that human oocyte spindles are surprisingly unstable\, and identified functions for actin and a liquid-like meiotic spindle domain for spindle assembly in mammalian oocytes. \nIn the main part of her talk\, she will present recent work from her lab that sheds light on the origin of high aneuploidy rates in mammalian embryos. The vast majority of human embryos are aneuploid. Aneuploidy frequently arises during the early mitotic divisions of the embryo\, but the origin of this remains elusive. Using bovine embryos as a model for human embryos\, we identify an error-prone mechanism of parental genome unification which often results in aneuploidy. Surprisingly\, genome unification initiates hours before breakdown of the two pronuclei that encapsulate the parental genomes. While still within intact pronuclei\, the parental genomes polarize towards each other\, in a process driven by centrosomes\, dynein\, and microtubules. The maternal and paternal chromosomes eventually cluster at the pronuclear interface\, in direct proximity to each other. Parental genome clustering often fails however\, leading to severe chromosome segregation errors\, incompatible with healthy embryo development. Nucleoli\, which associate with chromatin\, also cluster at the pronuclear interface in human zygotes. DESCRIPTION:“How to make microtubules and build the cytoskeleton”\nSabine Petry\, Ph.D.\nAssociate Professor\, Molecular Biology\nPrinceton University \nAbstract:\nHow does a cell construct its microtubule cytoskeleton? According to Feynman’s principle “what I cannot create\, I do not understand”\, my lab pursues this question by building the chromosome segregation machinery from scratch. I will first tell you how the microtubule framework is generated in a cell. Upon deciphering the function of the most important microtubule accessory proteins\, I will present how we use those building blocks to reconstitute a spindle substructure in vitro and determine its building plan. Finally\, I will outline how we combine spindle substructures like pieces of a puzzle to assemble and thereby understand a functioning spindle that segregates chromosomes. \nBy studying how the MT cytoskeleton is built\, I hope to help explain how hundreds of proteins can self-assemble on the nm scale into a complex molecular machine 1000-fold larger than its constituents\, a challenge for the biochemistry of the 21st century \n  \nPlease email mbiasst@lifesci.ucla.edu to request Zoom link.
